High Throughput Liquid Handling

The Target Discovery Institute high throughput screening facility employs two full integrated PerkinElmer Janus Liquid Handling Work Stations, BioTec Plate Washers, platestackers, and FlexDrop reagent dispensers for all screening liquid handling.

Janus Liquid Handing Workstation 1 is housed in a ScanLaf Class 1 biological robotics enclosure for cell based small compound and siRNA high throughput screening.

Janus Liquid Handing Workstation 2 is housed in a BigNeat Class 2 biological robotics enclosure for cell based small compound and RNAi including siRNA high throughput screening.

High content imaging

High Content Imaging

The Target Discovery Institute utilizes two high content imagers:

1. GE InCell 1000 High Content Imager is automated with a SCARA laboratory robot and controlled by PAA Overload 2 scheduling software to execute large batch cell based high content imaging screens.

2. PerkinElmer Operetta High Content Imageris automated with a SCARA laboratory robot and controlled by PAA Overload 2 scheduling software to execute large batch cell based high content imaging screens. There are several objectives and an extensive range of filters covering all commonly used fluorophore.

More information on High Content Imaging (including protocols and stains available) can be found here

Envision

High Throughput Plate Reader

PerkinElmer EnVision Multilabel Plate Reader is used at the Target Discovery Institute for plate reader applications and can be integrated directly with the liquid handling work stations or through automated plate stackers depending on the requirements of the screen. The EnVision plate reader is capable of performing AlphaScreens, Fluorescence Intensity, Fluorescence Polarization,Time-Resolved Fluorescence (TRF), luminescence and absorbance.

LSRII

High Throughput FACS

The Target Discovery Institute currently utilizes a BD LSRII with High-Throughput Sampler (HTS) module flow cytometer for FACS based high throughput screens located at The Experimental Medicine Division Flow Cytometry Facility based in Room 5605, Level 5, John Radcliffe Hospital.
